Thirlby was born in Cambridge, the first of three children. By 1521 he had graduated BCL from Trinity Hall, Cambridge. After his undergraduate years, he was elected a fellow of his college, and proceded to doctorates in civil and canon law in 1528 and 1530. He received patronage from Stephen Gardiner, the king's secretary who was also Master of Trinity Hall. From 1528-1531 Thirlby was working for the University, on legal affairs.
